While it is possible to control other toolbars using the built-in CUI tools, the toolbar for the relationships grid hasn't yet been migrated to use these tools. This means that it is currently only possible to make global changes that will be applied to all relationships by modifying files in the code tree.
If you are only looking to disable the lock button, you should be able to accomplish this by configuring the standard permission structure to limit your users' ability to edit these related items.

Yes, in recent service packs of 12.0, it is now possible to add a button to the relationship tab using CUI. You can check out this blog post for an overview of the most commonly used CUI locations.
